Материал и методы. Работа выполнена на трабекулах ушка правого предсердия пациентов с ИБС и больных ИБС, ассоциированной с СД2. Функцию СР оценивали по изменению показателей инотропной функции изолированных мышц в изометрическом режиме при выполнении теста с периодами покоя. Уровень Са2 +(АТФ(азы (SERCA2a) определяли методом Вестерн-блоттинга. Результаты. Обнаружено, что инотропная реакция на периоды покоя может быть двух типов. I тип характеризовался тем, что после периодов покоя амплитуда сокращений либо оставалась на уровне базовых значений в случае пациентов с ИБС, либо превышала базовые значения в случае больных с сочетанной патологией. При II типе реакции инотропный ответ мышечных полосок значительно угнетался в обеих группах пациентов. Миокард с I типом реакции характеризовался более высоким уровнем экспрессии Са2 +-АТФазы СР, при этом у пациентов с СД2 уровень SERCA2a был достоверно выше, чем в миокарде у пациентов без СД2. При II типе реакции, независимо от группы, экспрессия SERCA2a была значительно ниже, чем при l типе реакции. Заключение. Сочетанное развитие ИБС и СД2 с небольшим сроком заболевания характеризуется сохранением функций кальций-транспортирующих систем СР и более высоким уровнем SERCA2a в кардиомиоцитах, чем при изолированном развитии ИБС, что может иметь важное значение при определении лечения и прогноза. </p></abstract><trans-abstract xml:lang="en"><p>Aim. The aim of the study was to investigate the function of sarcoplasmic reticulum (SR) in cardiomyocytes from patients with ischemic heart disease (IHD) associated with type 2 diabetes mellitus (T2DM). Materials and Methods. The study was performed with right atrial appendage trabeculae from patients with IHD associated with T2DM. SR function was evaluated through changes in the inotropic function of the isolated muscles in isometric contraction regimen during the post(rest test. Ca2+-ATPase (SERCA2a) levels were evaluated by Western blotting. Results. Study showed the existence of two different types of inotropic reaction. The first type was characterized by a preservation of the post(rest twitch amplitude at basal levels in patients with IHD or by an increase in the amplitude in patients with IHD associated with T2DM. The second type was characterized by a significant decrease in the inotropic response in the both groups of patients. Concentration of SERCA2a was higher in the myocardium from patients with the first type of inotropic response, being the highest in patients with T2DM. The myocardium of patients with the second type of inotropic response had low concentration of SERCA2a, without a significant difference between patient groups. Conclusions.
